Exploring the relationship between gut microbes, phages, metabolites and cardiometabolic health using computational methods

Award details:

Metabolomics and the gut microbiome are intricately involved in the risk of metabolic diseases. Moreover, there is increasing evidence that metabolites produced by the gut microbiota, such as short-chain fatty acids, bile acids and branch-chain amino acids, are responsible for several metabolic conditions including insulin resistance, diabetes and obesity, thus mediating microbial effects on human health.  Likewise, the gut bacterial communities and their activities can be altered by viruses, including phages, making these biological entities important regulators of the host’s health. For instance, gut virome alterations have been associated with a wide range of diseases. However, the impact of the gut virome on the metabolome and health is still scarce. The objective of this PhD studentship is to identify the contribution of the gut microbiome, virome, and metabolome to cardio-metabolic disease and to investigate their interactions. This will provide a better understand the underlying mechanisms of phage-host interactions and their impact on the human host. 

The PhD student will analyse available data from TwinsUK, a register of UK twins, and from the post-prandial nutritional intervention study ZOE PREDICT-1. These two studies provide a wealth of existing data, including blood and faecal metabolomics, the gut microbiome (shotgun metagenome), detailed cardiometabolic phenotyping at multiple time points, dietary information and post-prandial responses. The virome analysis will be supervised in collaboration with Prof. Li Deng at the Technical University of Munich and Helmholtz Centre Munich in Germany.

Seeking highly motivated early career researchers with a strong statistical/mathematical/bio-informatics background for a PhD studentship in human metabolomics/microbiome at the Department of Twin Research & Genetic Epidemiology, King’s College London. The Department has close links with Divisional and other Research Departments within King’s College, as well as many external Research Organisations. Our group includes over 60 researchers and produces over 70 papers per year in high impact factor journals including Nature, Cell and Nature Genetics and are ranked in the world top 1% of cited scientists. The PhD student will also work in close collaboration with the Deng lab in Munich, that has developed an integrative approach combining single-cell technologies, culture-independent techniques, multi-omics, machine learning, as well as their developed pipeline called ViroProfiler (https://github.com/deng-lab/viroprofiler) to analyse viral metagenomic data.

The successful candidates should have at least a BSc or equivalent in the areas of bio-informatics, statistics or mathematical sciences. Title: EPSRC Studentship: Design of High-Performance Electrode Materials for Sustainable Non-Aqueous Aluminium-Ion Batteries (King’s College London Scholarships)

About the Project

The PhD project will focus on the development of innovative electrode materials for non-aqueous aluminium-ion batteries.

Background: Due to concerns over the environmental impact of lithium-ion batteries and the use of critical materials, alternative battery systems including non-aqueous aluminium-ion batteries (AIBs) have gained increased attention. AIBs are advantageous in that they contain highly abundant aluminium replacing critical lithium, have low cost, and a high theoretical energy. However, AIBs are still in their infancy and further research is needed to understand their detailed working principles and how to control the underlying reactions.

Project details: This project therefore aims to gain an in-depth understanding of the electrochemistry in AIBs and how to tune their performance. To this end, a systematic study of the impact of the chemical composition and materials engineering on the reaction mechanisms will be conducted. The obtained findings will be then rationalised into a design strategy for sustainable high-performance cathode materials for AIBs. Ultimately, the results of this project shall lead to the development of optimised AIB systems and will have a significant impact on the current battery landscape.

The student will use solid-state and solvothermal synthesis methods for the preparation of the electrode materials. In depth analysis of the materials is done via complementary characterisation techniques such as electrochemical cycling, X-ray diffraction, X-ray photoelectron spectroscopy, electron microscopy, atomic force microscopy and more. Further, a computational component can be added. In addition, also the environmental and economic implications of AIBs will be explored using life cycle assessment and techno-economic modelling.

Requirements: We are especially interested in students with a strong understanding of materials science, chemistry, chemical engineering, or relevant topics. Lab experience (chemical synthesis) is required. Additional knowledge in electrochemistry, energy storage systems and/or sustainability-related modelling is welcome but not a requirement.

This interdisciplinary project is positioned at the interface between materials science, engineering and sustainability and provides ample opportunities for internal and external collaborations.

The British Council Palestine and King’s College London hold an agreement to jointly support up to 2 Master’s Scholarships and 1 PhD Scholarship at King’s College London each year.

Background:

The ability of high-temperature superconducting (HTS) materials to carry large current densities with little or no resistance makes them attractive for providing large rotor magnetic fields in high-power density superconducting electrical machines. Superconducting electrical machines are more efficient, smaller and lighter than conventional ones, and there is great interest in exploiting these properties for next-generation electric aircraft and other electric transport applications to meet ambitious net zero emissions targets.

HTS flux pumps – which exploit the nonlinear resistivity of an HTS wire when subjected to a varying magnetic field – offer the possibility of a brushless exciter for a wire-wound superconducting machine to inject a large DC current into its rotor coils without the need for slip rings/brushes and thermally-inefficient current leads. This would significantly improve the efficiency and reliability of such machines.

Project details:

This project aims to develop state-of-the-art numerical models of the HTS dynamo-type flux pump, in order to design and optimise the device for use as a brushless exciter for superconducting electrical machines in electric aircraft. Thus, in addition, a thorough analysis of electric aircraft requirements will be required to set design specifications.

To validate the numerical models, the project will also involve proof-of-concept experiments, which will ultimately enable us to explore the scaling up of the device for real-world application.

The Ph.D. will involve part of, but not restricted to, the following topics:

1 – Exploring the mysteries of bio-inspired flight/swimming. More specifically, the candidate will use theoretical and computational techniques to model natural flight and swimming flow problems, understand the mechanism behind them and provide advanced control and optimization strategy for the bio-mimic systems. The candidate could get involved in the fundamental analytical analysis on vortex shedding and the role of vortices in incompressible flows, the CFD simulations on 2D and 3D bio-inspired flight/swimming cases, the fluid-structure-interaction analysis, the data-driven surrogated model through artificial intelligence (GNNs, RNNs, LSTM, etc.), or the advanced control strategy and optimization.

2 – Modelling, control, and optimization of the flow in renewable energy systems, specifically wind turbines/farms or proton exchange membrane (PEM) fuel cells. In more specific terms, the candidate will simulate the fluid or fluid-structure interaction problem in wind turbines/farms or the multi-disciplinary multi-scale problem in PEM fuel cells numerically. The candidate will also deliver the flexible modelling framework for PEMFCs and provide solutions for the next generation PEMFCs by comprehensive optimization in the aspects of flow channel design, cooling strategy, input gas ratio, etc., to achieve high efficiency, reliability, and long durability.

The candidate will work with Dr. Juan Li (first supervisor) and Dr. David Moxey (second supervisor) based at Strand Campus, King’s College London. Dr. Li is experienced in the field of unsteady aerodynamics and renewable energy. Dr. Moxey is an expert in developing cutting-edge novel numerical methods from academia and their application to challenging industrial fluid dynamics problems. To limit global warming to the target of 1.5°C, COP26 ended with a global agreement to accelerate action against climate change this decade alongside many countries aiming for longer-term goals of net zero emissions by 2050. The building sector emissions are historically one of the largest sectors when including direct and indirect emissions from heating, electricity and cooking it accounts for 28% of global emissions, followed closely by the transport sector with 27%. Approximately 60% of emissions from both of these sectors is from consumer energy demands, making personal energy use from residential buildings and cars around 33% of global emissions.

To address personal energy (e.g., heating/cooling, transport, and electricity) decarbonisation, previous studies usually focus on the interaction between these demands, with work completed primarily at national level. However, as energy demands, investment for personal energy decarbonisation is usually paid by customers. It is critical to allow an understanding of if low-carbon technologies will be viable for consumers, and therefore if they will be taken up at mass, which may lead to different results to that which is optimum by regional level long term analysis.

This project is to lead the development of numerical models and analysis tools for consumer energy decarbonisation within regional/global perspectives. Objectives are to 1) analyse domestic energy decarbonisation (electricity, heating and transport) options from the customer’s perspective (one region or across regions in the world), 2) assess if decaronisation will worsen equality of access to energy services (e.g., electricity, water, etc), and /or 3) assess if bottom-up customers’ least cost options are compatible with top-down system optimums. Also, like www.HeatMyHome.ninja project, this PhD project will develop modelling tools. The PhD project is funded by KCL for over 3.5 years.
